Research and Development of Living Plants in-vivo Fluorescence Imaging System for Experimental Teaching

Through the development of plant culture and imaging system, a low-cost plant living fluorescence imaging system was developed. The system adopts a double-layer box structure design, which is divided into two parts: the constant culture system and the fluorescence recording system. It can provide the necessary life support function for plant growth, and can also obtain the key images in the process of plant growth in real time, including the time-space mode of monitoring the fluorescence signal in the development of the transgenic plant. The EGFP transgenic tobacco and Arabidopsis were successfully cultured and observed in real time using this system. The low cost greatly expanded the application of the system in experimental teaching, and also laid a foundation for the further study of the observation and detection of follow-up life activities.

Whole Mount in situ Localization Technology of Plant Hormones

The existing phytohormones measurement methods are relatively cumbersome and difficult to measure the concentration of intracellular hormones. The paper describes whole-mount “immunolocalization” technique for the overall distribution of auxin (IAA) during lateral root and flower development in Arabidopsis. Based on the specific reaction of the antibody to the antigen, this method binds the antibody to the secondary antibody linked to enzymes, fluorescent molecules, etc. to detect the auxin in tissues or organs by light and electron microscopy, realizing the overall in situ localization of auxin (whole mount in situ localization of Hormone, WISIH). This method is not only suitable for growth hormone IAA, but also for rapid and reliable tissue or organ localization of hormones such as abscisic acid (ABA). The protocol is usually done within three to four days. It is a highly sensitive, specific and simple hormone in situ localization method, and is suitable for various plant tender organs and tissues.

Improvement of Experimental Techniques of Whole Mount in situ Hybridization Technology in Rice

Taking rice SUPERWOMAN1 (SPW1)/OsMADS16 gene as an example, by optimizing the preparation of hybridization probes, material immobilization, dissociation, permeabilization and color development, the SPW1/OsMADS16 gene expression patterns with high specificity but low backgrounds were obtained. The procedure of whole mount in situ hybridization (WISH) technology for rice is simple and inexpensive, and it can be performed simultaneously with several species in the centrifuge tube. The main experimental steps include probe preparation, material immobilization and dissociation, hybridization reaction, post-hybridization treatment and detection and color development. The proposed method provides a technological basement for a protentional high throughput detection of spatiotemporal gene expression patterns and functional analysis in rice.

Study on the Classification of Response Relationship between Total Pollutant Emission Reduction and Water Quality Improvement in China

 The response relationship between total pollution emission reduction and water quality improvement from 2011 to 2015 was analyzed by data envelopment analysis (DEA). The results show that the response relationship is different in time and space. The response relationship was further divided into four different models, and suggestions about total pollution emission controlling and water environment prevention were made under different models, in order to make total pollution emission controlling play a better role in water quality improvement. The result should provide scientific basis for precise pollution control in accordance with the water environment quality objectives of the basin.

Estimating Nitrogen and Phosphorus Pollution Load in Bali Lake Basin of Jiangxi Province Based on SWAT Model

The simulation of nitrogen (N) and phosphorus (P) pollution load structure in Bali Lake basin was performed by SWAT model using the spatial and attribute database acquired through social survey and statistical data. The model was applied and validated based on the data of meteorology, runoff and water quality from 2010 to 2014. The spatial-temporal distribution of N and P in the basin was also studied. The results showed that the urban life was the most important source of N and P load, following by agriculture and livestock breeding pollution source, and the least was industry pollution source. The annual TN and TP loads were mainly concentrated in flood season (from May to September) which took up nearly 55% of the whole year load. As the amount of precipitation increased, TN and TP loads increased during flood period, which was mainly due to the increasing of non-point rural living water and agricultural fertilization pollution source with scouring action of the heavy rainfall during flood season. The spatial distribution of N and P was centered on the Jiujiang county, Xunyang and Lushan district as the urban agglomeration area affected by urban sewage pollution.

A Novel Unicast Based Multiplexing Scheme to Improve the Capacity of VoWLAN

The mergence of VoIP and WLAN created a new wireless platform for voice service delivery, VoWLAN. This paper figured out the capacity bottleneck of VoWLAN, and analyzed the problems in current solutions such as M-M. A novel unicast based multiplexing scheme (U-M, Unicast Multiplexing) to improve the capacity and performance of VoWLAN was proposed. Both theoretical and simulation results showed that this scheme improved the voice capacity to nearly 100% comparing to the traditional un-multiplexed scheme, and reduced the packet lost ratio by 10% compared with the M-M scheme. Meanwhile, the U-M scheme is a power-saving scheme in the voice-oriented application as well. Therefore the proposed U-M scheme is an effective solution to increase the voice capacity of WLAN and improve the performance of VoIP service.

Calculational and Distributive Method of Urban Total Permitted Air Pollutant Emission

Total air pollutant emission control planning is a major urban environmental management approach. A set of technical schemes have been developed, which are applicable to study the calculational and distributive method of the total quantity control for urban air pollutant emission in China. The permitted total emission equivalent quantity of urban air pollutants and its distribution in urban area, which can be used as regional air pollution control index and can be allocated to each major pollution source as its limit of emission quantity, can be worked out by using linear programming model based on regional grid air pollutants transfer matrix which considers the difference of functional regions and the justice of the permitted emission's distribution. The application of the technical paths and models in Jinan, indicates that it is reasonable and feasible.

The psaC Gene of Synechococcus sp. PCC 7002 encodes the FA/FB aprotein of photosystem I, a protein with a molecular weight of 8.9 kD containing two 4Fe-4S centers. The expression vector of psaC gene was constructed, and transformed into Saccharoromyces cerevisiae.Southern blotting confirmed that psaC gene had been successfully transformed and Tris-Tricine-PAGE revealed that psaC gene had been highly expressed after incubating the transformed strains in the inducing culture.
