血液中异山梨醇[1, 4; 3, 6]二醚-5-单硝酸酯的测定

Determination of Isosorbide-5-mononitrate in Human Plasma

摘要: 建立了一种改进的简单、灵敏、准确地测定血液中异山梨醇[1, 4; 3, 6]二醚-5-单硝酸酯的毛细柱气相色谱方法。血浆样品加入饱和Na2CO3溶液,以乙醚作萃取剂,甲醇重溶解, 2, 4-二硝基甲苯作内标物,最低检出限可达5μg/L。标准曲线相关系数大于0.999。对高中低3种浓度血浆样品的日内和日间相对误差和相对标准偏差均小于8%。回收率92%-104%。

Abstract: An improved, sensitive and precise capillary gas chromatographic method with electron capture detector has been developed for the determination of isosorbide-5-mononitrate in human plasma. 2, 4-Dinatrotoluene is used as the internal standard. The plasma is extracted with ether in sodium carbonate solution. The residue was reconstructed with methanol. The linear correlation coefficient between concentration and height ratio is better than 0.999. The recovery was found to range from 92% to 104% for three concentration levels of samples with a limit of determination 0.000005g/L and with the relative standard deviation of 8% within-day.
