Abstract: Anti-IF-protein antibodies were used as probes for immuno-fluorescence, and the reactions of them with different parts of the cell were detected, which suggested the possibility of the existence of the intermediate-like filaments in the cytoplasm. Those proteins homologous to the antibodies distributed regularly in the protoplasm. To characterize the corresponding proteins, SDS-PAGE and immunoblots were utilized. The 21, 23, 33 and 68kD proteins were distinguished among the diverse protein constituents of the cell. Some of these proteins also showed the cross-reactivities with anti-IF-proteins antibodies derived from higher organisms. These two evidences both contributed to the homology of some proteins in Paramecium to the intermediate filament proteins in higher organisms.

摘要: 以抗中间纤维蛋白抗体作为探针进行免疫荧光实验,得到细胞内不同部位的阳性反应,暗示原生质中可能存在类中间纤维。这些同源蛋白的胞内分布具有一定的规律性。进一步采用SDS-PAGE和免疫印迹技术研究它们的生化性质,发现4种主要蛋白明显有别于胞内其他蛋白组分。它们是21,23,33及68kD蛋白。在免疫印迹实验中,4种蛋白中的一种或几种分别与不同的中间纤维蛋白抗体有交叉反应。以上实验初步表明,草履虫细胞内存在高等生物中间纤维蛋白的同源蛋白。
