Effects of Partially Purified Neurotoxins from Scorpion Buthus Martensii on the Sodium Currents of GH3 Cells

Abstract: Two partially purified neurotoxins, peak II and III, were obtained from the crude venom of scorpion Buthus martensii by fractionation on Sephadex G-50. Bioassay experiments on cockroaches and mice indicated that peak II was more toxic to cockroaches and less to mice, while peak III was just reversed. The effects of these two partially purified neurotoxins on sodium currents of rat anterior pituitary cells (GH3 cells) were investigatedusing the whole-cell patch clamp technique. Peak III induced a pronounced slowing of Na+ inactivation, which is a typical response to a α type anti-mammal toxin from the Old World scorpion venom. Peak II dramatically shifted the voltage dependence sodium currents activation to much more negetive potentials, just like a typical β type anti-mammal toxin from the New World scorpion. It is interesting to see that a partially purified anti-insect toxin from the Old World scorpion behaved as a β type anti-mammal toxin on mammalian sodium channels.

摘要: 马氏钳蝎粗毒经Sephadex G-50柱层析分离后可得到几个组分。其中峰Ⅱ毒素对蜚蠊毒性较大,而峰Ⅲ毒素对小白鼠毒性较大,可以认为它们是部分纯化的昆虫类毒素和哺乳类毒素。用膜片箝全细胞记录方式研究了峰Ⅱ、峰Ⅲ毒素对大鼠垂体前叶克隆化细胞GH3细胞钠电流的影响,结果表明峰Ⅲ毒素使钠电流失活变慢,是典型的α型哺乳类毒素;而峰Ⅱ毒素使电压依赖性钠激活曲线显著地向负的方向移动。这种部分纯化的昆虫类毒素对一种哺乳动物细胞钠电流的影响表现出典型的β型哺乳类毒素的性质。
