    Mineralization-Alteration and Genesis of Lejiawan Antimony Deposit

    Lejiawan antimony deposit is a typical deposit in the antimony mineralization zone in northern Guang Dong Province. Zoning of mineralization and alteration in Lejiawan antimony deposit is very distinct, from orebody to country rock there are antimony mineralization-belt, silification-belt and pyritization-belt, respectively. Based on the investigations and analyses of the ore-forming process, isotopic data and fluid inclusion data, the authors suggest that the genesis of Lejiawan antimony deposit belongs to stratabound epithermal transformation deposit.

    Mouse metallothioneins-I genomic gene was inserted into pSV2-dhfr plasmid at the sites of EcoRI and the recombinant plasmid was transferred into CHO-dhfr-cells by DNA-calcium phosphate coprecitation method. Using western Blotting, RNA Hybridization, we could find the expression of MT-Ⅰat the level of 1.8μg/106 cells in D-22 clonal cells which were selected by DMEM selective medium more than 8 weeks.The D-22 cells were treated with cisplatin and the IC50 to cispiatin was 0.145 μmol/L, being 3.1-fold resistance to cisplatin compared with the nontransfected cells—CHO-dhfr-cells (0.04 μmol/L). Theseresults provided the evidence of that Metallothionein may play an important role in protecting CHO cells from cytotoxicity of cisplatin. So increasing thesynthesis of MT by the inducer such as BSN and blocking the synthesis of MT bythe inhibitor such as proparglycine may be a potential way to treat some refractory carcinomas in chemotherapy.

    A peptide (35kD) was purified by gel filteration with sephadex LH20 and SDS-PAGE from storage proteins of wheat. The peptide is a material which can be used to form peptide vesicles after being sonicated in some aqueous solvent. The peptide vesicles can wrap various compounds of molecular weight ranging from 0.4 to 150 kD. The peptide vesicles which wrap the compounds can be kept in stable state for months at 4℃ or for days at room temperature. The vesicles can be changed into scale state by freeze drying and scale state can be changed into state of vesicles by rehydrating. That is to say, they have the characterization of D R F (dehydration rehydration vesicles). The peptide vesicles prepared from modified protein can wrap water-soluble compounds, but they can only wrap hydrophobic compounds if the protein is not modified. It shows that the hydrophobic region is located inside membrane of the peptide vesicles. The protein contains 16 hydrophobic amino acid residues and a N-terminal Phe.

    Herbicide-resistant transgenic upland rice plants were recovered using biolistic method. Upland rice suspension cells and immature embryos were transformed by particle bombardment with a plasmid containing herbicide resistant bar gene. One week after bombardment, the bombarded cells were transferred to N6 medium with phosphinothricin (PPT) for selection. Fertile transgenic plants were regenerated from PPT-resistant calli. Integration of bar gene was confirmed by Southern blot hybridization. Resistance of R0 transgenic plants to herbicide PPT was shown by Basta (commercial form of PPT) applications in the greenhouse. Inheritance of bar gene was demonstrated by PCR analysis of R1 transgenic plants.

    Experiments were conducted during April-Oct. 1994 in Beijing rice field. Largecultivar differences in methane emission flax have been found. Variety 93812 emitted about fivefold more CH4 per meter square per hour than did the variety Qiuguang. Organic amendment plus (NH4)2SO4 as the mixed base fertilizer and (NH4)2SO4 as the topdressing applied in different amounts and stages compared with the mixed base fertilizer only could reduce methane emission about 58% and increase rice yield about 31.7%. Emission Peaks of CH4 in the tillering stage and reproductive stage could be obviously suppressed.

    A chitosancontaining glassy carbon electrode was found to give highly sensitive voltammetric wave for Pd(Ⅱ)-KCl-HCl system by anodic stripping voltammetry. Various conditions for the determination of trace Pd(Ⅱ) were studied. The optimum chitosan amount in glassy carbon was 1% 4 μL. In a solution of 0.02 mol/L KCl-HCl(pH=2), at 0.0 V, electrodeposited for 2 min or 1 min, scan rate was 0.1 V/s, the anodic stripping peak potential was at +0.67 V(vs.SCE). The peak current is proportional to the Pd(Ⅱ) concentration between 0.075-9.9 mg/L, r=0.999 3. 1.5 μg/L Pd(Ⅱ) was determined by electrodeposition for 10 min. The method was applied to the determination of palladium in catalyst. The possible mechanism of electrochemical reaction has been proposed by study of CV and UV.

    L-tyrosine perchlorate was synthesized in aueous solution. Its crystal structure was determined by X-ray diffraction method. The single crystals belong to monoclinic, space group P21 with a=0.5335(1) nm, b=0.9817(2) nm, c=1.200 0(3) nm,β=94.79(2)0,V=0.6263 (3) nm3,Z=2,Dc=1.494g/cm3.

    A retroviral vector pINS-GH carrying human growth hormone gene (hGH) was introduced into murine embryonic stem (ES) cells. Aithough expression of hGH gene was not detectable by radioimmunoassay, Southern pattern showed that hGH gene did integrated into genome. The ability of differentiation of transfected ES cells were tested in vitro and in vivo. The result indicated that ES cells also remained their pluripotency after a series of manipulation. Two chimaeric mice have been obtained by blastocysts injection of the transfected cell line GC-5.

    The charge transfer interactions of dobutamine with p-benzoquinone and chloranil were investigated in ethanol medium, the reactions of Dobutamine with p-benzoquinone and chloranil are complete. The products have maximum absorption at 500 nm and 472 nm, respectively. The apparent molar absorptivities are 1.81×104 for p-penzoquinone and 4.22×104 for chloranil. The method proposed on the basis of the above results was applied to the determination of Dobutamine injection and the results were satisfatory.

    The solution structures of cryptand (2,2) and its Lanthanum nitrate cryptate intheir acetonitrile solution were studied, basing on their 1H, 13C, 1H-1H and 1H-13C NMR (Nuclear Magnetic Resonance) spectra. The cryptand has several distinct conformations in solution which are in equilibra of rapid interconversion. It also possesses the same symmetry as that of its crystal structure, For Lanthanum nitrate cryptate, its conformations are fixed to some extent by the action of coordination of La3+. It is deformed and rigid as well. However, it still keeps its original symmetry. By the method of heavy water exchange, we obtained the proton chemical shift of NH groupin cryptand (2,2).

    Dongfengshan gold deposit is occured in hornstone of Dongfengshan group under ironbearing stratum. Ore-bearing stratum contain richly yrrhotite,pyrite,spessartine and dannemorite. Additionly, there are smaller arsenopyrite and cobaltite. The δ34 S values of pyrrhotite and pyrite are -15.4‰~9.8‰ and -2.7‰~6.1‰,respectively. Sulfur of sulfide occurrence mainly from plutonic sourse. The δ18 O values of different rocks and minerals show following: marble 13.1‰~23.9‰, magnetite 4.1‰~10.5‰, quarts 13.7‰~20.9‰, grunerite 10.9‰~16.8‰, garnet 9.9‰~17.5‰. These δ18 O values reflect thermal contact metamorphic character of gold deposit .The δ18 O values of the Paleozoic granite obviously are of two groups:one is terrene granite in southern ore area ,its δ18 O values are 8.7‰~11.4‰;another is drill holl plutonic granite in northern ore area, its δ18 O values are 0.5‰~1.1‰. The granite from drill holl was strongly sericitized. Dongfengshan group was formed in early Proterozoic. Dongfengshan gold deposit is a high temperature thermal contact metamorphic syngenetic deposit.

    Bile acids play an important role in the digest process. It was found that various bile acids exhibited different biological and spectroscopic behavior. We supposed the difference may be correlated with the different capability of forming intra-molecular hydrogen bond. So the possibility of forming intramolecular hydrogen bond for various bile acids were investigated by molecular mechanics calculation code MM3. In this paper, we proposed a new pre-optimized calculation method, which was used along with the MM3 molecular mechanics calculation to evaluate the possibility of forming intra-molecular hydrogen bond in various bile acids. The calculation results show that (1) 17R-7α-OH-bile acid is not able to form stable intra-molecular hydrogen bond while the 17S-7α-OH-bile acid can form two stable intramolecular hydrogen bonds. The stability of the conformation is proved by the fact that the conformation has lower steric energy and no negative vibration frequncies. (2) 17S-12α-OH-bile acid can also form intra-molecular hydrogen bond.

    A hybridomas secreting McAb 1G10 which can inhibit the catalytic activity of carboxypeptidase A (CPA) was prepared and characterized. Analyzing the model of cell nucleus, main chromosome number was between 80-90. The titer of the ascites fluid was 106. Binding constant for McAb 1G10 to CPA was 1.59×109(mol/L)-1. McAb 1G10 was identified as belonging to the IgG1 subclass.Its molecular weight was about 150 Kd. The interaction of McAb 1G10 with CPA was investigated. A competitive inhibition of peptidase activity of CPA by McAb 1G10 was shown by Dixon Plot. The apparent inhibitory constant obtained by this procedure was 1.17×10-7 mol/L. These results suggested that the binding site of McAb 1G10 with CPA may be the active site of CPA for its peptidase activity. Based on this work, the idiotypic antibody which is the “Internal-Image” of CPA can be prepared and may have the similar enzymatic activity with CPA.

    By means of micronucleus test of bone marrow cell and sperm malformyion test, the protective effects of the Cordyceps sinensis(Berk) Sacc, artificial pypha on genetic damage induced by cigarette smoke was studied in Kunming mice stain. The results show that mutagenic potential in groups treated with cigarette smoke for 1, 2 and 4 weeks was noticed. The antimutation function of the Cordyceps sinensis (Berk) Sacc, artificial pypha is very evident in groups for 2 weeks. The rate of suppression micronucleus is 55%. No significant difference was found among the percentage of sperm malformtion of two treating groups (P>0.05).

    Isolated hearts tacken from nonhibernating ground squrrel in Autumn were perfused by Langendorff method with Krebs-Henseleit solution. Hearts were first perfused for 10 min to establish steady state, then stopped for 15 min ischemia (noperfusion), and finally followed by 10 min reperfusion. Coronary effluents were sampled at certain time intervals during noperfusion and reperfusion period. The enzymatic activity of CK(U·L-1) was analysed from the coronary effluent, in the mean time the ECG and contraction amplitude were measured. Parallel experiments with the same protocal have been performed in nonhibernator animal, rat, for comparasion. During 15 min noperfusion period there was a remarkable CK release from both animal hearts. The amount of CK release was proportional to the noperfusion time course in both hearts, but the extent of CK release form rat heartwas much more bigger than that of ground squirrel. During reperfusion period the CK release of rat heart was biphasic while that of the ground squirrel was monophasic and the peak of the 1st releasing phase of the former (255±17 U·L-1)was much higher than that of the later (179±32U·L-1). The incidence of ventricular fibrillation and ventricular tachycadia as well as the percentage of decreasedcontraction amplitude were all significantly reduced in ground squirrel heart in comparation with rat heart. These results indicate that the heart from ground squirrel exhibits a stronger resistence to ischemiareperfusion injury than that from rat, presenting a striking contrast between hibernator and nonhibernator.
    Isolation and Purification of Metallothionein from Liver and Intestine of Giant Salamander Induced by Cadmium

    The giant salamander is very interesting animal from evolutionary point of view,because its transitional position from aquatic to terrestrial life, therefore, the present investigation dealing with the metallothionein (MT) molecular evolution of the giant salamander provides significant clues to evolution of vertebrate. Each extract of liver and intestine of giant salamander induced by subcutaneous injection of cadmium (CdCl2) was separated by gel filtration chromatography Sephadex G-50 column,then was separated into twoisometallothioneins (isoMTs) by ion-exchange chromatography DEAE Sepharose Fast Flow column. According to the results of gel filtration HPLC detection, the molecular weight of the two isoMTs was about 12000 dalton. The isoMTs contained Cd and Zn (cadmium and zinc) and the value of Cd/Zn was about 3/1. The Cys content was about 21%~24% and the product amount were about 628μg·MT/g·wet liver, 185μg·MT/g·wet intestine. The isoMTs have the typical amino acid composition of metallothionein and U.V. absorption shoulder near 250 nm.
