Effects of Recombinant Human Trefoil Factor 3 on the Cell Proliferation and Migration

Abstract: To investigate the mechanism of trefoil factor 3 on the gastric intestine epithelial restitution, the recombinant human trefoil factor 3 was added to human colonic tumor (HCT) cell and the proliferation effect was examined by MTT assay. The recombinant protein didn't promote the proliferation of the HCT cells at low density of 0.01~0.05 g/L and only has weakly proliferation effect at density of 0.1~0.2 g/L. 1 g/L of the recombinant protein could significantly promote the cell migration of HCT cells when added to the monolayers cells.

摘要: 将重组人三叶因子3(Trefoil factor 3, TFF3)作用于人结肠肿瘤细胞,研究重组蛋白对细胞增殖的影响,结果发现该蛋白在较低的浓度(10~50 mg/L)下对细胞的增殖基本没有影响,在100~200 mg/L浓度下该蛋白对细胞仅有微弱的刺激作用,提高浓度对细胞增殖作用没有改变。同时研究了TFF3对损伤的单层结肠肿瘤细胞迁移的影响,发现TFF3对细胞有明显的促进迁移作用。
