Study on the Reaction between Proteins and the Complex of Cu(II)-sulfochlorophenol S and Its Application

Cu(Ⅱ)-氯磺酚S配合物与蛋白质反应及分析应用的研究

Abstract: Cu(II)-sulfochlorophenol S could combine with BSA to form a blue three component complex in Clark-Lubs buffer at pH 2.5, which possesses maximum absorption at 724nm with 53, 93 and 160nm of red shift compared to the Cu(II)-sulfochlorophenol S complex, BSA-sulfochlorophenol S complex and dye itself, respectively. The standard curves of some kinds of proteins were constructed under optimum condition obtained in our lab. The linear range is large. The assay is sensitive, accuracy, and tolerant to many foreign substances. Moreover, all the reagents are stable and the performance is easy to carry out.

摘要: 用染料探针对蛋白质的定量已应用于临床医学及生命科学的研究中,但染料以金属配合物的形式对蛋白质的定量及其机理研究较少。本文首次报道,在pH 2.5的 Clark-Lubs 缓冲介质中,用氯磺酚 S-Cu(Ⅱ) 配合物与BSA作用,生成深蓝色的三元复合物,该复合物的λmax为724nm,比其金属配合物的λmax红移53nm,比试剂本身红移160nm, 比氯磺酚S-BSA的二元复合物红移93nm。研究了最佳反应条件,制作了几种蛋白质的工作曲线。测定的线性范围宽,灵敏度高。用于实际人血清样品的测定干扰少,准确度高。
