Establishment and Characterization of Rabbit ES-like Cell Lines

兔ES样细胞系的建立及其特性分析

Abstract: Seven continous ES-like cell lines were established from 237 rabbit embryos. 90 and 96 hours embryos were plated on ME (mouse embryonic fibroblast cells) feeder layer, using DMEM/F12 medium with 10% FBS and 10% rabbit serum supplemented with LIF or EGF. ES-like cells isolated from these embryos shared many characteristics with mouse ES cells: morphology of stem cell, colonial growth, maintenance of an undifferentiated state through serial passage, ability to differentiate spontaneously or by induction, ability to form embryoid bodies in suspension culture. Using these cells as nucleid donor, nuclear transfer resulted in 11% blastocyst development.

摘要: 报道从237枚家兔胚胎中建成7个可连续传代的ES样细胞系。建系条件为,使用小鼠原始胚胎成纤维细胞(ME)作饲养层,以含10%胎牛血清和10%兔血清的DMEM/F12为培养基,添加白血病抑制因子(LIF)或上皮生长因子(EGF),胚龄为90,96h。该细胞系的细胞,在许多方面类似于小鼠ES细胞:具干细胞的形态特征,呈集落型生长,可连续传代并保持其形态特征,具一定的自发分化和诱导分化的能力,悬浮培养可形成简单类胚,但皮下接种同种乳兔未获肿瘤。用所建的兔ES样细胞进行卵母细胞核移植时,有11%发育为囊胚。
